AIE-Based CRISPR-Cas Applications

The integration of aggregation-induced emission (AIE) technology with CRISPR-Cas systems is a major advance. This chapter explores the latest applications of CRISPR-Cas based on AIE luminogens (AIEgens) in the fields of biosensing and gene editing. Known for their bright fluorescence when aggregated, AIEgens utilize their unique optical properties as signal probes that combine the precision recognition, cis-cleavage activity, and multiple turnover trans-cleavage activity of the Cas system to amplify the output signals of biosensor platforms. The synergy has facilitated the development of highly sensitive biosensors capable of detecting trace amounts of biomarkers. Moreover, the Cas system also has the ability to efficiently knock out genes. AIEgens can also help accurately monitor the gene editing process of the Cas system and enhance its imaging and therapeutic evaluation capabilities. Finally, this chapter briefly outlines future opportunities for AIE-based CRISPR-Cas. The combination of AIE with CRISPR-Cas technology not only enhances the capabilities of biosensing and gene editing but also paves the way for further research and clinical applications, potentially enabling more precise and safer gene manipulation technologies. This advancement is anticipated to lead to a new era of improved molecular tools in biomedical research and clinical applications.
