Polymeric Microsponges for Drug Delivery to Ocular Candidiasis Infections: Preclinical and Clinical Evidence

In this chapter, we explore the latest advancements in microsponge delivery systems (MS-DS) for ocular drug delivery, along with their potential applications in combating ocular fungal infections, particularly candidiasis. The eye provides a unique challenge for drug delivery due to its protective barriers, rapid tear turnover, and limited residence time. Microsponges offer a promising approach by enhancing drug retention, providing controlled release, and improving corneal penetration, thereby ensuring sustained antifungal activity. Given that fungi thrive in moist environments like the conjunctiva and corneal surface, ocular candidiasis remains a serious concern, often leading to severe complications if untreated. Conventional antifungal treatments struggle with low bioavailability and frequent dosing requirements, which microsponge-based formulations can effectively overcome. Additionally, this chapter provides an in-depth review of microsponge fabrication techniques, with a special focus on the quasi-emulsion solvent diffusion method, a widely adopted approach for developing ophthalmic drug carriers. We further discuss the advantages, challenges, and future prospects of microsponge-based ocular drug delivery systems in treating fungal infections and other ophthalmic disorders.
