DNA methylation is a powerful epigenetic marker with growing applications in forensic science, particularly for body fluid identification (BFID). This chapter explores the role of forensic DNA methylation in distinguishing biological fluids, beginning with an overview of bisulfite and enzymatic conversion techniques used to analyze methylation patterns. It then highlights advancements in detection methods, such as massively parallel sequencing and methylation-sensitive PCR, which enable precise methylation profiling. Additionally, the chapter discusses the development of predictive models, the integration of DNA methylation with SNP typing for enhanced forensic DNA profiling, and emerging approaches for deconvoluting mixtures. As forensic epigenetics continues to evolve, DNA methylation is poised to become a crucial tool for improving forensic investigations and evidence interpretation.
