The Y-chromosome offers great utility to the field of forensic DNA analysis. The Y-chromosome has been subject to forensic use for over 30 years, particularly in cases involving sexual assault, paternity, mass disasters, and other case types in which autosomal DNA fails to yield useful results. Methods such as Y-screening, Y-STR analysis, and RM Y-STR analysis can be used to determine the most probative items of evidence in sexual assault cases, generate investigative leads, ascertain kinship, assist with human identification, and offer vital information regarding male contributors to DNA mixtures. This chapter describes the genetic construction of the Y-chromosome, its research history and relevance to forensic DNA analysis, and the use of Y-screening and Y-STRs in typical forensic casework.
