Yeasts are unicellular eukaryote classified as members of fungus kingdom. They are important members of the probiotics that play essential immunomodulation roles in aquatic animals. In this study, two yeasts were isolated from mangrove leaves in Songkhla province, southern Thailand, namely MUHT042 and MURM012. Molecular identification by PCR and phylogenetic tree analyses was conducted using the oligonucleotide primers, FA2 (5´GTCTGGTGCCAGCAGCCGCG3´) and RA2 (5´CCCGTGTTGAGTCAAATTAAG3´) specific to the partial nuclear small subunit 18S rRNA region. The obtained sequences were compared with reference sequences in the GenBank database using the bioinformatic tools, BLAST, and were subsequently used for construction of a maximum-likelihood phylogenetic tree by MEGA11 software. The bootstrap support was evaluated with 1000 replicates. Based on the 18S ribosomal RNA sequences, the MUHT042 and MURM012 isolates obtained in the present study were identified as Rhodotorula paludigena and Rhodotorula mucilaginosa, respectively. The phylogenetic tree with highest log likelihood of R. paludigena and R. mucilaginosa was found using the Hasegawa-Kishino-Yano model when compared with Jukes-Cantor, Kimura 2-parameter, Tamura 3-parameter, Tamura-Nei and General Time Reversible models. The present study indicated that bioinformatic techniques are useful for identification of yeast samples.
