LYTAC: Lysosome-Targeting Chimeras

The LYTAC (lysosome-targeting chimera) technology was first proposed in 2020 by Carolyn R. Bertozzi and others [1]. LYTAC is a new therapeutic strategy that induces the degradation of disease-related proteins in the lysosomes within cells. LYTAC is a chimeric molecule that combines a ligand, such as an antibody, which specifically binds to target proteins on the cell surface or outside the cell, and a ligand that binds to proteins involved in shuttle transport to the lysosome, forcing the target protein to be transported to the lysosome for degradation (Fig. 113.1). This technology enables the removal of cell surface or extracellular proteins that were difficult to approach with conventional drugs and is expected to be used as a protein degradation drug.
