Commercialized Pharmaceutical Implants and In Situ Forming Depots Using the Polar Aprotic Solvent DMSO

Dimethyl sulfoxide (DMSO), an apolar protic solvent, has been used in parenteral applications since the 1960s. It is an active pharmaceutical, a processing aid, and as an excipient in multiple, commercial long-acting injectable products. The ability of DMSO to dissolve several biodegradable polymers such as poly(lactide-co-glycolide) makes it an attractive solvent for extended-release injectable products. Care should be taken when using DMSO in processes that use plastic parts such as tubing and gaskets as well as final primary and secondary packaging. Evaluating processes and conducting stability studies in the early stages of development are recommended.
