Stem cells offer an array of advantages over conventional drug therapies since they are inherently biomimetic and offer numerous beneficial mechanisms and factors involved in tissue repair. Stem cells, particularly mesenchymal stem cells (MSCs) have emerged as promising therapeutics for salivary gland regeneration owing to their higher regenerative potential and less immunogenicity. Over the past 10 years, due to the success of several pre-clinical studies, including modifications of somatic cells to have controlled potency of stemness and tissue regeneration, these findings have progressed to clinical trials. Numerous studies using pre-clinical animal models show MSCs as an effective treatment modality for salivary gland dysfunctions. In this chapter, we cover the recent pre-clinical and clinical studies involving adult MSCs for salivary gland regeneration conducted from 2015 to 2025.
