As carriers of lipids in the blood circulation, lipoproteins are instrumental to lipid homeostasis in the body. Decades of research on their contribution to cardiovascular health have revealed that lipoproteins can interact with target cells – including immune cells – via receptors. Through their interactions with cells, lipoproteins participate in the modulation of cellular metabolism and function. However, much has yet to be uncovered on the impact of lipoproteins on immune cells in the tumor microenvironment (TME). Immune cells are heavily present in the TME, and their activity can either promote or hinder cancer progression. In light of the ability of nanoparticles to address drug bioavailability, target-specific drug delivery and tumor penetration challenges, several studies have shown that HDL-type nanoparticles are well-suited for targeting of cancer cells as well as immune cells. In this chapter, we focus on the role of high-density lipoprotein (HDL) in cancer progression and in the functional phenotype of immune cells of the TME. In addition, this chapter provides an overview on the application of HDL-type nanoparticles in cancer immunotherapy.
