The Platelet Lipidome

Lipids both mediate and modulate platelet response as they serve as an energy resource and act as extracellular cues that trigger platelets into either action or inactivity. Lipids channel messages as signalling intermediates, instigate intraplatelet calcium mobilisation and signal ageing, apoptosis or procoagulant potential on assorted pathophysiological demands. Recent advancements in lipidomics analyses have been a boon for platelet biologists in unravelling intricate molecular perspectives from platelet biogenesis to functionality, providing disease-relevant biomarkers and devising stratagems for potential therapeutic interventions. Current state-of-the-art lipidomics analysis has evolved through improvements in instrumentation, offering better sensitivity and resolution, expansion in the availability of internal lipid standards for accurate annotation and precise quantification, complemented by robust development in systems biology tools, lipid databases and search engines that enable us to charter the vast territory of platelet lipidome. Platelet lipidomics has emerged as an essential tool in bridging experimental research and clinical studies for translational applications; therefore, regular appraisals and improvisations are required. This chapter recapitulates recent lipidomics insights from platelet biogenesis, (immuno)thrombosis, thrombo-inflammation and atherothrombotic cardiovascular disease and elaborates on the intriguing contribution of lipids as targets and mediators of therapeutic intervention in diseases necessitating administration of antiplatelet drugs.
