Antibiotics that inhibit synthesis of nucleic acids including DNA and RNA are presented. Background biochemistry information on structure of DNA, replication, and transcription is provided. Antibiotics discussed include DNA intercalators, topoisomerase inhibitors such as nalidixic acid and fluoroquinolones and nitroheterocycles such as nitroimidazoles and nitrofurans, RNA synthesis inhibitors such as actinomycin D and rifamycins are also included. Mechanisms of action of these drugs and resistance development against them are discussed.
