Cancer Metabolism

Metabolism is essential for the maintenance and propagation of life. Maintenance metabolism provides homeostasis for non-proliferating cells, whereas proliferating and cancer cells require a switch to proliferative metabolism that is controlled by proto-oncogenes and tumor suppressor genes. Deregulation of oncogenes or loss of tumor suppressors renders tumor cells proliferative in a fashion that they are dependent on nutrients and are hence vulnerable to metabolic inhibition. Because cells of the tumor microenvironment, particularly immune cells, also depend on metabolism, the therapeutic window for metabolic therapy needs further definition for developing new drugs.
