Endophytic actinomycete metabolites offer potential for medical applications, serving as a significant source of antimicrobial compounds to combat antibiotic-resistant microorganisms. Zingiber officinale Rosc. is common in Vietnam, especially in Ho Chi Minh City, renowned for its diverse array of bioactive compounds and antimicrobial properties. In this study, endophytic actinobacteria were isolated from Zingiber officinale Rosc. and screened for their antibacterial activity. Roots of Zingiber officinale Rosc. were surface-sterilized, then cultured on inorganic salt starch agar (ISP4) and actinomycete isolation agar (AIA). Subculture was performed to obtain pure strains. Isolates with actinomycetes-like morphology observed via Gram-staining were selected and cultured in broth for ethyl acetate extraction, then the antimicrobial activity of the extract was evaluated using agar-well diffusion testing. Finally, strains with high antimicrobial activity were subjected to identification using 16S rRNA sequencing. As a result, thirty isolates having different morphology under Gram-staining were retrieved from the two selective media. Seven isolates presented antimicrobial activity via agar-well diffusion testing. Isolate 4.7 and 4.11 had activity against Staphylococcus aureus, isolate 4.11 against Escherichia coli, isolate 4.5 and 4.7 against Bacillus subtilis, and isolate 4.6 and 4.9 against Pseudomonas aeruginosa. Among seven isolates identified with 16S rRNA sequencing, four were Streptomyces griseorubiginosus and three were Microbispora hainanensis. In conclusion, thirty endophytic actinomycetes-like isolates were isolated from the root of Zingiber officinale Rosc. Among those, seven isolates showed moderate to high antimicrobial activity. Seven isolates with high antimicrobial activity were identified, belonging to Streptomyces griseorubiginosus and Microbispora hainanensis.
