Aims: The study aimed to evaluate the effect of G-CSF treatment in bile duct ligated (BDL) mice pathology and its isolated hepatocyte. Method: The mice were injected with 5 consecutive days of G-CSF (61.5 μg/kg/day), and the mice were sacrificed to evaluate results after G-CSF injected 2 days and 14 days. Results: Normal mice injected with G-CSF show a significant decrease in the mRNA expression level of Il-1β, Il-33, Csf3r, and Hnf4α genes compared with the control group at 2 days post-operation (p < 0.05). Despite the downregulation of the onecut1 gene (encode for Hnf6) in the normal-treated model group, the difference was non-significant. In the BDL model group, gene expression of Il-1β was elevated in the BDL placebo group and significantly reduced in BDL + G-CSF (p < 0.05). There was no significant difference between the two groups for Il-33 and Csf3r expression (non-significant). In contrast, the Hnf4α gene increased its expression in the BDL-treated model group compared with the control group on the 26th day after surgery (p < 0.05). We recorded the uptrend of onecut1 gene expression in both BDL groups compared with the control (non-significant). Conclusion: G-CSF reduces the mRNA expression of the interleukin 1 subfamily, Csf3r, Hnf4α on normal-mice model hepatocytes. In the evaluation of BDL-treated G-CSF hepatocytes, the result shows that the mRNA expression of IL-1β was downregulated, while Hnf4α rose, and Csf3r remained consistent throughout the experiment. This result showed no statistical difference in the hepatocyte’s mRNA expression of IL-33 and onecut1.
