Application of Hexaploid Bread Wheat SNP Arrays for the Genotyping of Tetraploid Durum Wheat

Genotyping of tetraploid durum wheat (Triticum turgidum L. ssp. durum, genomes A, B) is currently performed mainly by using single nucleotide polymorphism (SNP) markers. For large-scale parallel genotyping of numerous SNP markers during research and breeding applications, the predominant technology is the use of arrays. Because of the relatively lower significance and smaller-scale breeding efforts for tetraploid durum wheat compared to hexaploid wheat, no comprehensive SNP array development has been performed, specifically for tetraploid wheat. Rather, many different genotyping arrays developed for hexaploid wheat (Triticum aestivum L., genomes A, B, D) have also been used for durum wheat. In this review, we describe some important issues in using hexaploid wheat genotyping arrays for the analysis of durum wheat. Specifically, we describe the differences in marker characterization and scoring, effects regarding the analysis of D genome markers with respect to functionality and polymorphism, and provide examples regarding the use of hexaploid wheat arrays in actual durum wheat breeding and research. Finally, we provide an outlook on possible future developments in SNP genotyping for tetraploid durum wheat based on DNA sequencing technologies.
