2-Naphthylamine is formed during the pyrolysis of organic substances containing nitrogen and as such can occur in nature. 2-NA has been detected in hair dyes as well as rubber antioxidants at levels of 0.25%. Several dye and pigment intermediates, paperboards, have been shown to contain small amounts of 2-naphthylamine. The main sources of exposure of the general population are from the environment, 2-naphthylamine and 2-nitronaphthalene, tobacco smoke, fumes from cooking oil, lubricants for automobile engines, in brake fluids, oils, contact with some types of dyes, and hair dyes contaminated with 2-NA. 2-Naphthylamine causes bladder cancer in humans, especially in Caucasians. Its urinary metabolites are 2-naphthylamine, N-(2-naphthyl)-hydroxylamine, bis-(2-amino1-naphthyl) phosphate, 2-aminobiphenyl, and 4-aminobiphenyl.
