Pan-JAK inhibitors, which inhibit multiple JAK isoforms (JAK1, JAK2, JAK3, and TYK2), offer a broad mechanism of action by modulating cytokine signaling pathways involved in skin inflammation. These agents have demonstrated efficacy in conditions such as atopic dermatitis, alopecia areata, vitiligo, and psoriasis, with some achieving rapid symptom relief and improved disease control. Their oral and topical formulations provide versatile options for treatment, although safety profiles, including risks of infections and hematologic abnormalities, warrant careful monitoring. Ongoing clinical trials and real-world studies continue to elucidate the therapeutic potential and longterm safety of pan-JAK inhibitors in dermatologic practice, positioning them as valuable tools in the management of challenging skin diseases.
