Skin Homeostasis Through AHR-Mediated Interactions Between Host and Microbiome

The microbiome, including the cutaneous microbiome, has been a center of attention since the commencement of the Human Microbiome Project, well over 15 years ago. The goal was to characterize the inhabitants of all body sites and to analyze the microbiome’s role in health and disease. Around the same time, the aryl hydrocarbon receptor (AHR) was identified to play an important role in epithelial barrier organogenesis, homeostasis, and maintenance. Evidence is emerging that the cutaneous microbiome can reciprocally interact with keratinocytes in the epidermis through activation of host’s AHR. Via host AHR signaling and downstream initiation of host defense mechanisms, the commensal inhabitants of the skin aim to protect their micromilieu and compete for their individual niche. This chapter aims to summarize the current evidence for physiological functions of microbe-driven, canonical AHR signaling in the skin and to set the stage for investigative studies on the AHR-driven molecular mechanisms of dermatological conditions in which microbiome dysbiosis is eminent. We identify opportunities to translate knowledge gained in other epithelial organs to physiological processes important in skin homeostasis.
