Electrical Bioimpedance Spectroscopy System Based on AD5933EBZ to Determine DNA Concentrations

Estimation of Deoxyribonucleic Acid (DNA) concentrations is a fundamental procedure in many molecular biology studies, requiring the use of specialized equipment. For instance, UV-Vis spectrophotometers, not always available in resource-limited settings. The electronegative nature of DNA molecules and viscosity related to the quantity of double helix molecules are expected to alter the volumetric electrical properties and bioimpedance magnitude of the samples. In this study, the use of the EVAL-AD5933EBZ evaluation board is proposed as a low-cost, easy-to-use technique for the determination of DNA concentration based on Electrical Bioimpedance Spectroscopy (EBiS). Results indicate sensitive EBiS magnitude values to differentiate three DNA concentrations ranges with relevance in molecular biology assays (<10, 10–25, >25 ng/μL), within a frequency bandwidth of 5 kHz to 50 kHz. EBiS measurements through a straightforward device as the AD5933EBZ seem to hold potential value to develop a cost-effective tool for estimating DNA concentrations. Further research is needed to assess the sensitivity and specificity of the AD5933EBZ limits for detecting different DNA concentration values.
