Biopesticides based on RNA interference (RNAi) are being developed as alternatives to conventional insecticides. RNAi biopesticides are highly specific and represent a new mode of action with improved safety profiles compared to synthetic chemicals. The first registered and commercialized foliar-applied dsRNA product, Calantha™, active ingredient ledprona, controls the Colorado potato beetle (Leptinotarsa decemlineata; CPB), a devastating pest of potato. Calantha™ was registered in the USA in 2023. Ledprona acts by specifically downregulating the expression of an essential gene in CPB while presenting negligible risks to non-target organisms or human health. Commercial-scale production of Calantha™ is enabled by a GreenLight Biosciences’ proprietary cell-free production process. Extensive field testing of Calantha™ demonstrated crop protection and potato yields equivalent to industry standard conventional insecticides. Calantha™ fits grower practices for ground, aerial, or chemigation application and is compatible with other pesticides and fertilizers commonly used in potato production. Recommendations for insecticide resistance management (IRM) in potato grower programs will help to mitigate against the chances of resistant CPB populations developing and safeguard the long-term effectiveness of Calantha™. Label recommendations limit the number of sprays to a single generation of CPB followed by rotation with an insecticide with a different mode of action. Calantha™ represents the first realization of a sprayable RNAi-based bioinsecticide, providing a paradigm for the development and registration of future dsRNA products.
