The Migratory Locust as a Model for Studying the Mechanisms of RNAi

RNA interference (RNAi) is a phenomenon of gene silencing triggered by short/small interfering RNA (siRNA) and found to function in most living organisms. Injecting double-stranded RNA (dsRNA) into migratory locust (Locusta migratoria) is an efficient and systematic method of RNAi, which can be used as an excellent model to study the mechanism of insect RNAi. In this chapter, we discussed the current research progress on various mechanisms related to L. migratoria RNAi efficiency, including dsRNA degradation, dsRNA cellular uptake and endosome escape, and RNAi core machinery enzymes. Additionally, we also discussed the use of symbiotic bacteria and fungi-mediated RNAi and dsRNA nanoparticles for pest management. We hope that the knowledge and methods developed by various RNAi studies using L. migratoria as a model insect will contribute to the RNAi research of other insect species and to the development of RNAi-based pest management strategies.
