RNAi in Chelicerata, Spider, and Mites: Meta-Analysis and Current Status of Two Model Organisms and Protocols

RNA interference (RNAi), also known as post-transcriptional gene silencing (PTGS), is a conserved mechanism for regulating gene expression in eukaryotes. Double-stranded RNA (dsRNA) is a key component in inducing RNAi. Today RNAi is one of the emerging genetic engineering techniques to effectively silence or inhibit the expression of target genes and it can also be used as a novel insecticide mode of action. This chapter describes the current status of RNAi in two important groups in the Chelicerata, a subphylum of the Arthropoda, which contains many important pests in agriculture but also in human health. After a concise introduction and a meta-analysis of the RNAi literature in Chelicerata (search in Web of Science with papers between 2000 and 2024), we present the current status of RNAi in two respective models, namely the pond wolf spider, Pardosa pseudoannulata, and the two-spotted red spider mite, Tetranychus urticae. In addition, we provide an easy, practical and efficient, step-by-step protocol for RNAi bioassays in spiders and spider mites, via injection and also via feeding, which has the potential to be widely applied. We hope this chapter will help to fill in the knowledge gap about RNAi in Chelicerates and also invite students and researchers who work on dsRNA in Arthropods.
