Histology of Salivary Glands

Major salivary glands, namely parotid, submandibular, and sublingual glands produce about 90% of saliva, and the remaining 10% is secreted by minor salivary glands. The major salivary glands have essentially similar microscopic structures. Each gland is wrapped in a fibrous connective tissue (capsule) that extends internally (septum) to divide the gland into lobes and lobules. The gland parenchyma is comprised of secretory end-pieces (acinus), intercalated ducts, striated ducts, and excretory ducts. Each acinus consists of several secretory cells, either serous or mucous cells. Acini of the parotid glands are almost entirely serous, while submandibular and sublingual glands have both serous and mucous acini, thus the latter glands are recognized as mixed secretion glands. Except for the lingual gland, each gland pours its secretion into the oral cavity via an excretory duct. Sublingual gland secretion flows directly into the oral cavity through multiple tiny ducts. The minor salivary glands are mainly submucosal mucous acini collections scattered around the oral cavity that release their secretion via tiny excretory ducts on the mucosa surface. Histologically, the minor acini are surrounded by loose connective tissue and have similar acinar cell structures to the major gland acini. Secretions of the minor salivary glands are mainly mucous or seromucous in nature except for von Ebner’s glands that present close to the base of the circumvallate papillae of the tongue, which are purely serous.
