Animal Amyloidosis

This chapter includes the processing of different animal amyloid deposits with topo-optical and immunohistochemical reactions. Confocal laser-scanning microscopic studies were also performed for this study. Both frozen and embedded material from different animals—dogs, cats, martens, cattle, mice, chickens and geese—were examined. Positive immunohistochemical reactions were seen in one case of AA amyloid in the spleen, pancreas, intestine and liver. We performed the topo-optical reactions described in Chaps. 3 and 4 and observed positive reactions in all cases. After HE staining, we observed amyloid birefringence in the preparates in polarized light. Using the CEC method, negative reactions were seen in goose and chicken amyloid preparates; i.e. keratan sulphate and heparan sulphate components were absent. After pretreatment with potassium permanganate or performiate, AA amyloid deposits lost their anisotropy, whereas AIAAP, fibrinogen and Aapo A-II amyloid deposits were resistant. After combined treatments, AA amyloid deposits were digested, whereas AIAAP, fibrinogen and Aapo A-II amyloid deposits became resistant. At the end of the chapter, we conclude with a question: What is the frequency of amyloidosis in animals?
