To address the limited understanding of polysaccharide biosynthesis in medicinal macrofungi, we present an integrated multi-omics protocol. Our method details the entire workflow, from experimental design and de novo genome assembly to transcriptomic and metabolomic profiling. Key genes are pinpointed through weighted gene co-expression network analysis and KEGG pathway mapping, followed by experimental validation using RT-qPCR. This established pipeline provides a robust framework for elucidating biosynthetic pathways and guiding strain improvement.
