Three-Dimensional Co-immunodetection and RNA-Fluorescence In Situ Hybridization in Preimplantation Mouse Embryos

We describe how to perform simultaneous immunodetection using fluorescence and RNA-fluorescence in situ hybridization (immuno-RNA-FISH). The aim of the approach is to localize proteins involved in epigenetic regulation (histone modifications) using immunodetection and the mRNA of genes of interest using RNA-FISH. This article describes an optimized protocol for mouse embryos that has been adapted for this specific biological material to preserve the three-dimensional (3D) organization of cell nuclei.
