Fluorescence In Situ Hybridization on DNA Fibers: Molecular Combing

Molecular combing is an advanced method for preparing extended DNA fibers. It enables the uniform and parallel stretching of hundreds of DNA molecules on a glass surface with an unprecedented resolution of two kilobase pairs per micrometer. In conjunction with fluorescence in situ hybridization (FISH), molecular combing enables direct visualization of genome structure and copy number variation, as well as quantification of sizes of overlap and gap between sequence contigs in genome assembly. Here, we present a multicolor fiber FISH protocol using DNA fibers produced by molecular combing.
