Light-sheet fluorescence microscopy has become a very popular method for imaging biological specimens. The very first implementation of a microscope based on planar illumination was the ultramicroscope, invented at the beginning of the twentieth century. Starting from this first instrument, this chapter reviews the history and development of light-sheet microscopy, up to the most recent advancements.
