While conventional methods focus on separating soluble from insoluble fractions, a significant part of bioactive compounds remains locked within cell walls. Harsher acid or alkaline treatments can free these compounds by loosening the rigid wall matrix. In addition, enzymatic or ethanol-based pretreatments can enhance extraction efficiency and purity. This chapter offers practical guidance on pretreatment, extraction, and purification of insoluble mushroom polysaccharides, with emphasis on reproducible protocols and improved solubility.
