Northern hybridization analysis of transgenic plants is done to confirm the presence of the transgene-derived transcript. Additionally, information on the size of the complete transcript and its relative expression can also be obtained by this technique. Presence of multiple forms (if any) of a transcript can also be visualized using this method. Radioactive labelling of DNA probe for Northern hybridization results in sensitive detection of the transcript. An elaborate protocol of Northern blotting and hybridization for detecting desired RNA from transgenic rice plants is described in this chapter.
