Best Practices for Protein Expression in Pichia pastoris Combining Design of Experiments with an Automated Platform for Parallelized Cultivation in Small-Scale Stirred Tank Bioreactors

This chapter provides practical guidance for studying the expression of proteins in the yeast Komagataella phaffii by using a commercial automated platform for parallelized cultivation in single-use stirred tank bioreactors. In addition to discussing the advantages and disadvantages of such a platform regarding scale, automatability, and single-use vessels, the main focus is on the presentation of a flexibly adaptable, operator-centric workflow for the use of the cultivation system. In particular, it includes highlighting typical pitfalls and sources of error in handling and culminates in the presentation of best practices to support the proper execution of such experiments. To complement, a generic guide to its use for strain characterization and process development using statistical design of experiments is suggested based on the parallelization capabilities of the system.
