In planta transformation strategies aid in the development of transgenics in difficult-to-regenerate or recalcitrant plant species. Many economically important crops including the legume pigeonpea fall under the group of plants that are less amenable to tissue culture. The strategy essentially involves in planta inoculation of apical meristems of germinating seeds and allowing them to grow into seedlings ex vitro. Since T0 plants are chimeric in nature, transformants are selected in the T1 generation. In this chapter, we present a shoot apical meristem-targeted in planta transformation in pigeonpea for resistance to pod borer, Helicoverpa armigera. This protocol can be extrapolated to other plant species as well as for the transformation of genes governing other traits.
