Biancaea sappan (L.) Tod., commonly known as Sappan wood or Pathimukha, is a notable species valued for its medicinal properties. This flowering tree flourishes in tropical regions and has traditional uses in rural communities. In India, the heartwood of Biancaea sappan is included in toothpaste due to its effective healing properties, particularly in stopping gum bleeding. Biancaea sappan is widely distributed across Southeast Asia. Its dried hardwood serves both as a natural coloring agent in foods and beverages and as a home remedy for various health issues. Cryopreservation has proven to be a safe and cost-effective method for preserving germplasm and managing in vitro-produced materials for biotechnological purposes. Specifically, the cryopreservation of pollen has emerged as an effective strategy for germplasm conservation. This chapter outlines a standardized protocol for the cryopreservation of Biancaea sappan pollen, while also discussing its significance in conservation efforts and utilization within breeding programs.
