Crinum species, belonging to the family Amaryllidaceae, are ornamental plants valued not only for their aesthetic appeal but also for their medicinal significance, owing to the presence of bioactive alkaloids with neurological and anticancer properties. These plants play a crucial role in traditional medicine, horticulture, and ecological stability. However, habitat loss, climate change, and restricted distributions threaten their survival, necessitating urgent conservation efforts. Cryopreservation of pollen grains presents a reliable method for conserving genetic material, thereby supporting hybridization, research, and conservation initiatives by ensuring year-round availability and facilitating breeding programs. This chapter outlines a cryopreservation protocol for five Crinum species native to India—Crinum asiaticum L., C. latifolium L., C. lorifolium Roxb., C. malabaricum Lekhak & S.R.Yadav, and C. viviparum R.Ansari & V.J.Nair. Detailed procedures for pollen collection, viability assessment, dehydration, cryogenic storage, and post-thaw viability evaluation through in vitro germination are provided. The establishment of a cryo bank for Crinum germplasm contributes to the long-term conservation and sustainable utilization of these ecologically and economically significant species.
