Cryopreservation is an efficient and reliable tool for the long-term conservation of cashew (Anacardium occidentale) genetic diversity. The significant variability within the cashew family makes it a valuable genetic resource for both conservation and crop improvement programs. Preserving plant genetic resources (PGR) is crucial for safeguarding valuable genetic diversity that is increasingly threatened by biotic and abiotic stresses. Cryopreservation offers a promising solution to conserve cashew biodiversity effectively. In this study, pollen grains were collected from male flowers between 9:30 and 11:30 A.M. and assessed for viability using the in vitro germination hanging drop technique. The collected pollen was subsequently dried under light, packed, and stored in liquid nitrogen. In vitro and in vivo pollen viability was evaluated by conducting crosses under field conditions using cryopreserved pollen. This chapter outlines the cryopreservation protocol for cashew pollen, including dehydration and subsequent storage in liquid nitrogen.
