The analytical technique of choice for the separation, identification, and quantification of carotenoids is the high-performance liquid chromatography (HPLC) with UV/Vis detection. Here, we describe basic procedures for the preparation and extraction of carotenoids in human blood and faecal samples and their analysis using C18 and C30 columns. The methods detailed in this chapter describe the type and volume of extraction solvent used, the duration of the successive steps, and the identification and quantification system and some considerations for obtaining the best parameters of recovery, selectivity, and specificity.
