Photolabile linkers are chemically very stable and can be cleaved under mild conditions without destructive reactions on target molecules. o-Nitrobenzyl-based linkers are widely used in solid phase synthesis. Recently, our group developed several o-nitrobenzylic-based photolabile resins retaining different modifications at reducing-end of glycans after photocleavage. Here, we described the synthesis of three different o-nitrobenzylic derivatives and the immobilization of linkers on commercial Merrifield resin. Subsequently, detailed procedures of loading determination and photocleavage are presented.
