Sulfonopeptides are sulfur analogues of naturally occurring peptides and have been widely applied as inhibitors of certain enzymes due to their tetrahedronal structural feature, which is a mimic structures of transition states in the amide and ester hydrolysis. Here the synthetic method of sulfonopeptides is described. The method focuses on the widely utilized coupling of the corresponding sulfonyl chlorides and amino acid/peptide esters.
