Loop-Mediated Isothermal Amplification (LAMP): A Simple Method for the Detection of Foodborne Pathogens

Loop-mediated isothermal amplification (LAMP) is a molecular diagnostic tool that has been exponentially implemented in the diagnostics field, especially during and after the SARS-CoV-2 pandemic. LAMP enables exponential amplification of any nucleic acid targets of interest at a set temperature facilitating the use of simple heating devices. Due to the nature of how the nucleotide sequence is amplified, LAMP can produce a positive signal in under 30 minutes and exhibits increased sensitivity compared to the standard DNA amplification techniques based on Polymerase Chain Reaction (PCR). This includes techniques that are commonly used in diagnostics such as real-time quantitative (qPCR) and digital droplet PCR. Here we summarize the different methodologies and technical considerations for setting a LAMP diagnostic assay for foodborne pathogen detection. The methods can be utilized to amplify a DNA or RNA target with minimal equipment and reagents, with the view to highlighting its application for detecting foodborne pathogens in poor resource settings.
