Protocol for Isolation of Lung Fibroblasts from Mouse Tissue

This protocol describes the process for isolating lung fibroblasts from mouse tissue, which are commonly used in studies of lung biology, fibrosis, and regenerative medicine. Lung fibroblasts play a critical role in the maintenance of lung architecture and the response to injury, making them valuable for research into pulmonary diseases. The procedure involves harvesting a lung lobe from an euthanized mouse via thoracotomy, followed by enzymatic digestion with HyqTase enzyme to dissociate the tissue. After several washing and centrifugation steps, the fibroblasts are cultured in a suitable medium, where they begin to migrate from the tissue fragments. This method provides a reliable approach to isolating and cultivating fibroblasts for further experimental analysis, including studies of cell behavior, gene expression, and drug responses.
