Base editing (BE) is a powerful and efficient tool to introduce a designed base substitution in the genome. Several BE variants have now been developed that differ in base-change pattern, editing window, protospacer adjacent motifs (PAMs), and off-target effects. One of the first and foundational base editing tools developed in our laboratory was Target-AID, which efficiently performs C to T base substitutions at specified positions and has been successfully applied to the genome modification of plants including rice and tomato. This chapter first summarizes current plant applications of various base editing technologies, followed by a detailed protocol for Target-AID in rice.
