Gel electrophoresis is a widely used analytical method for protein characterization. It also serves as a foundational step for various other analytical techniques such as Western blotting, N-terminal amino acid sequencing, and mass spectrometry. Within this chapter, we describe the protocols for vertical protein electrophoresis using nondenaturing, non-dissociating polyacrylamide gel electrophoresis (NDND-PAGE), and sodium dodecyl sulfate (SDS)-PAGE. Furthermore, procedures are outlined for sample preparation, gel preparation, and band visualization through Coomassie Brilliant Blue staining, silver staining, and glycoprotein staining.
