Bacterial engineering through transformation techniques is a useful tool for studying plant–microbe interactions by tagging bacterial strains with fluorescent proteins. In this chapter, we describe the electroporation technique, a physical method that induces the acquisition of extracellular DNA by bacterial cells. In particular, we present a well-set-up electroporation protocol for Gram-negative bacteria that includes a double selection for the detection of transformant cells.
