Genetic information of plant microbiome is an important approach to explore the relationship between plants and microorganisms. PCR amplification and high-throughput sequencing technology can more accurately reveal the species and genetic diversity of microbial communities. In this chapter, we extracted plant genomes using the modified CTAB method, PCR amplification of plant endophytic fungi using 18S and ITS region primers, and high-throughput sequencing and analysis. Through this method, the information of microbial community structure of endophytic fungi allowed us to obtain.
