The most important methods for the determination of protein concentrations are UV absorption, biuret formation (with amplifying reactions for increased sensitivity), dye binding and amine-reactive reagents. The results are relative (colour is as intense as given by a x mg/mL solution of reference protein y). Depending on the method used and the reference protein chosen, the resulting concentrations for a given protein sample may vary by a factor of 20!
