The purpose of enzyme kinetics is to elucidate the mechanism of enzymatic reactions. To this end the reaction velocity is determined at different concentrations of substrates and inhibitors and under different environmental conditions (pH, temperature, pressure, etc.). Enzyme kinetics is not directly interested in the physiology of the enzyme or its substrate(s). Hence, the reaction conditions chosen may well be unphysiological, as long as they yield information on mechanism.
