An important step in the characterisation of a new protein is the determination of its primary structure (amino acid sequence). This can be determined either by repeated chemical cleavage of terminal amino acids (either from the \(\mathsf {N}\) - or \(\mathsf {C}\) -terminal end), or by MS/MS.
