Culture, Characterization, Differentiation, Lentiviral Transfection, and Functional Analysis of Human Bone Marrow-Derived Mesenchymal Stem Cells to Pacemaker-Like Cells

Human bone marrow-derived mesenchymal stem cells (hBM-MSCs) represent a valuable source for regenerative medicine due to their multipotency and ease of expansion in vitro. This protocol describes a comprehensive workflow encompassing the culture and maintenance of hBM-MSCs, their phenotypic validation via immunocytochemistry and flow cytometry, trilineage differentiation, lentiviral transfection, functional analysis through digital polymerase chain reaction (PCR), and cardiomyocyte-like functional validation using an impedance-based real-time cell analysis system.
