Adoptive cell therapy (ACT) is an approach for the separation and reinfusion of autologous or allogeneic cells. Non-genetically modified or genetically modified cells expressing new chimeric antigen receptors such as T cells, natural killer (NK) cells, or engineered macrophages are among the most interesting types. This technique is considered a promising method to treat cancer in patients with solid tumors, resulting in long-lasting responses and, in some circumstances, even a cure in certain situations. However, the ACT has limitations to be used in the treatment of malignancies because the tumors are weakly antigenic; as a result, tumor-specific T cells which have affinity and specificity for the antigens survive in the patient after chemotherapy. The short-term survival of CTLs in the body with no presence of antigen-specific Th cells and cytokine infusions is another restriction of the adoptive transfer of CTLs. Recent advancements in cell-manufacturing techniques have the potential to establish cell treatments as viable therapeutic choices in the future. In this chapter, we spoke about the different types of immune cells used in adoptive cell therapy, successful clinical trials and progress that has been done, and the limitations of each chosen immune cell.
