Synthesis and Antiarrhythmic Activity of New 1,3,4-Oxadiazole Derivatives Containing a 1,4-Benzodioxane Fragment

Abstract
The reaction of previously reported 4-(1,4-benzodioxan-6-yl)tetrahydro-2H-pyran-4-carbonyl chloride with hydrazine hydrate gave symmetrical diacylhydrazine which was cyclized to 2,5-bis[4-(1,4-benzodioxan-6-yl)tetrahydro-2H-pyran-4-yl]-1,3,4-oxadiazole by the action of phosphoryl chloride. Unsymmetrical 2,5-disubstituted 1,3,4-oxadiazoles were synthesized by the condensation of 4-(1,4-benzodioxan-6-yl)tetrahydro-2H-pyran-4-carbonyl chloride with various acid hydrazides, followed by cyclization of the resulting diacylhydrazines with phosphoryl chloride. The synthesized compounds were evaluated for their antiarrhythmic activity.
